A study of cotyledon senescence in cucumber (Cucumis sativus L.) based on expressed sequence tags and gene expression

Abstract: Cucumber cotyledons provide an excellent experimental system in which to investigate developmental changes in gene expression, from the early phase of heterotrophism through phototrophic growth to senescence. A cDNA library was prepared from the final stage of senescing cucumber cotyledons (>95% yellow) for studying the genes responsible for lipid mobilization during germination and senescence. This library had produced numerous senescence-associated clones in a previous study. Here, a total of 365 cDNA clones… Show more

“…Cucumber cotyledons offer a potentially ideal model system for this type of experiment, since one cotyledon leaf of a plant could be subjected to mineral analysis while the second one is harvested for RNA isolation. ESTs from senescing cucumber cotyledons have been described (Kim 2004), and with the completion of the cucumber genome (Huang et al 2009) genome-wide transcriptional profiling experiments are now feasible.…”

“…Previously, we proposed that the cucumber PEPCK and ICL genes function in senescing the cotyledon for recycling of the demolished intracellular organelles and supply their nutrients to other parts of the cell [6,8]. However, a different mechanism was proposed for ICL and PEPCK enzyme involvement in senescing organs that also needs to be elucidated in the future [1].…”

“…Genes and bioinformatics analysis Cucumber SAGs were presented in Table 1 been isolated from previous study [19]. However, we have cloned more than hundreds of cDNA clones which should be further identified the possible biological functions from the bioinformatics study through National Center for Biotechnology Information (NCBI) (Data not shown).…”

“…Polymerase chain reaction (PCR) was performed for 30 cycles using MyCycler (BioRad, USA) with an AccuPower PCR PreMix Kit (Bioneer, Korea) by gene-specific primers (5'-3'), as described in a previous report [19]. All primers were manufactured through Bioneer Co (Daejeon, Korea).…”
