A study of autophagy in hemocytes of the Pacific oyster, Crassostrea gigas

Abstract: Macroautophagy is a mechanism that is involved in various cellular processes, including cellular homeostasis and innate immunity. This pathway has been described in organisms ranging in complexity from yeasts to mammals, and recent results indicate that it occurs in the mantle of the Pacific oyster, Crassostrea gigas. However, the autophagy pathway has never been explored in the hemocytes of C. gigas, which are the main effectors of its immune system and thus play a key role in the defence of the Pacific oyste… Show more

“…Last but not least, it is noticeable that the pattern of autophagy indices was nearly uniform in both tissues used for this study; however, the differences were more distinct and significant in hemocytes than in gut cells. This corresponds to the recommendation of Picot et al 50 , who verified the usefulness of hemocytes of the oyster Crassostrea gigas under induction and inhibition conditions. They also explained this result by the fact that hemocytes play a crucial role in animal immunity and defense against pathogens.…”

“…This observation was further validated by CYTO-ID staining (Fig. 1C)(Picot et al, 2019). Next, we assayed autophagy maturation (fusion of the autophagosome with lysosome) in differentiating monocytes by DQ Red BSA staining(Zhang et al, 2012).…”

“…This finding was further corroborated by CYTO-ID staining (Fig.7C, S7E). CYTO-ID Autophagy Detection Kit labels theautophagic vacuoles and monitors autophagic flux in live cells using a novel dye that selectively labels accumulated autophagic vacuoles [47] [48].Next, we evaluated the autophagy maturation in stimulatedmonocytes.Thefusion ofautophagosomes with lysosomes is termed autophagy maturation. So to assayautophagy maturation, we used DQ Red BSA, a fluorogenic substrate for acid-resistant proteases.…”
