A Study of 5-Fluorouracil Desorption from Mesoporous Silica by RP-UHPLC

Abstract: In cancer treatment, the safe delivery of the drug to the target tissue is an important task. 5-fluorouracil (5-FU), the well-known anticancer drug, was encapsulated into the pores of unmodified mesoporous silica SBA-15, as well as silica modified with 3-aminopropyl and cyclohexyl groups. The drug release studies were performed in two different media, in a simulated gastric fluid (pH = 2) and in a simulated body fluid (pH = 7) by RP-UHPLC. The simple and rapid RP-UHPLC method for quantitative determination of … Show more

“…Highly specific articles both present fluorimetric studies on the crystal structure of the new polymorphic form of 3-aminoflavone [3] and the investigations of the dihydrogen bond in the aminoborane complex of a nicergoline intermediate [4]. Sulekova et al published a study evaluating the release of 5-fluorouracil from unmodified and modified mesoporous silica matrices [5]. A complex synthesis–analytical–galenical study described the synthesis of several piperazine-2,5-dione derivatives and their transdermal enhancement activity using theophylline as a model drug, and the prepared compounds can be potentially used as excipients in semi-solid drug formulations or transdermal therapeutic systems [6].…”

“…The optimal wavelength of 5-FU absorption using UV was at 204 and 266 nm by measuring the standard 5-FU solution in a sodium acetate buffer solution for 5-FU concentrations of 1-10 ppm [39]. We chose 266 nm to be the most optimum wavelength because the absorption spectra near 200 nm were absorbed by atmospheric oxygen that could affect the accuracy of the measured absorbance [40]. 5-FU solution at concentrations of 1, 2, 4, 6, 8, and 10 ppm was used to identify the standard linear curve.…”

“…Other than this poor nutrition, human papilloma virus infection, genetic factors, and oral hygiene will be a threat to the cause of oral cancer [6].Current treatments incorporate chemotherapy, radiotherapy and chemically derived drugs, Pallative Treatment and Surgery. Drugs which are used most often in chemotherapy are Cisplatin [7], Carboplatin [8], 5-Fluorourasil (5-FU) [9], Paclitaxel (Taxol) [10], Docetaxel (Taxotere) [11], Hydroxyurea [12], Methotrexate [13], Bleomycin [14] and Capecitabine [15]. In any case, these treatments have slumped in numerous perspectives making human life despondent and dwindling the life expectancy of patients.…”
