A Study of 11-[3H]-Tetrodotoxin Absorption, Distribution, Metabolism and Excretion (ADME) in Adult Sprague-Dawley Rats

Abstract: Tetrodotoxin (TTX) is a powerful sodium channel blocker that in low doses can safely relieve severe pain. Studying the absorption, distribution, metabolism and excretion (ADME) of TTX is challenging given the extremely low lethal dose. We conducted radiolabeled ADME studies in Sprague-Dawley rats. After a single dose of 6 µg/(16 µCi/kg) 11-[ 3 H]TTX, pharmacokinetics of plasma total radioactivity were similar in male and female rats. Maximum radioactivity (5.56 ng Eq./mL) was reached in 10 min. [ 3 H]TTX was b… Show more

“…administration. Meanwhile, TTX has also been shown to be rapidly distributed in the gastrointestinal tract after entering the body [ 20 ], suggesting that sodium channels in the gastrointestinal tract are blocked, which likely prolonged the duration of TTX absorption. This may be related to the long-term efficacy of oral administration.…”

“…On the other hand, metabolite analysis of plasma, urine and feces samples demonstrated the presence of only oxidized TTX. As a conclusion, TTX was found to be rapidly absorbed and excreted in rats [62].…”

“…administration (4.12 ng/mL). The value of V d was 192,752.13 ± 46,492.16 mL/kg, suggesting a low plasma concentrations of TTX, and TTX might mainly distributed in tissue or be confined in a certain organ, properly in intestine according to our previous study [5]. Compared with the results of i.v.…”

“…Further, the half-life (t 1/2 ) of TTX is short, and the effective blood concentration can only be maintained for about 2 h after i.m. injection [5]. In order to maintain the efficacy, frequent i.m.…”
