A standardized flow cytometric method for screening paroxysmal nocturnal haemoglobinuria (PNH) measuring CD55 and CD59 expression on erythrocytes and granulocytes

Abstract: PNH is a disorder of the pluripotent stem cells resulting in a deficient expression of membrane-bound GPI-anchored proteins in different cell types. Several flow cytometric approaches are designed to detect this antigen deficiency. But they all require drawing and testing of normal samples as control. Therefore, in the present study two flow cytometric assays for the detection of CD55 and CD59 deficiency in erythrocytes (REDQUANT CD55/CD59) and granulocytes (CELLQUANT CD55/CD59) are proposed. Precalibrated bea… Show more

“…23 The screening of paroxysmal nocturnal hemoglobinuria clones was carried out by flow cytometric analysis of CD55 and CD59 expression in erythrocytes, and glycosyl-phosphatidylinositol expression in neutrophils with cutoff values of 3.0% and 0.1%, respectively. 24,25 Pretransplant comorbidities of patients were assessed and categorized according to hematopoietic cell transplantation-specific comorbidity index. 26 Neutrophil and platelet engraftment was defined as an ANC ⩾ 0.5 × 10 9 /L on the first 3 consecutive days and a platelet count ⩾ 20 × 10 9 /L without transfusion support on the first 7 consecutive days, respectively.…”

“…Both commercially available and clinically validated kits REDQUANT (Biocytex, Marseille, France) and CELL-QUANT (Biocytex, Marseille, France) were used for the evaluation of CD55 and CD59 expression of erythrocytes and granulocytes according to the manufacturer's instructions [16]. Furthermore, FLAER combined with multiparameter flow cytometry (CD45, CD33, and CD14) in parallel was used for the detection of GPI-deficient PNH clones (granulocytes) as described previously [17].…”

“…The extent and the character of platelet dysfunction, underlying mechanisms, and involved cascades of platelet activation processes including receptor activation, platelet adhesion, clot formation under low and high shear rates, and aggregation remain unclear. Therefore, we have decided to use a broad panel of platelet assays including global methods as well as receptor-directed flow cytometric assays 24,25 to analyze platelet function combined with the quantitation of the PNH clone size in platelets and other peripheral blood cells. 26 …”
