A Spheroid‐Forming Hybrid Gold Nanostructure Platform That Electrochemically Detects Anticancer Effects of Curcumin in a Multicellular Brain Cancer Model

Abstract: In this study, a multifunctional platform that enables the highly efficient formation of 3D multicellular cancer spheroids and precise real‐time assessments of the anticancer effects of curcumin in a brain tumor coculture model is reported. A highly conductive gold nanostructure (HCGN) is fabricated to facilitate cancer spheroid formation without using anti‐cell adhesion molecules. A neuroblastoma (SH‐SY5Y) and glioblastoma (U‐87MG) coculture model is generated on HCGN with a specific cell‐to‐cell ratio (SH‐SY… Show more

“…Figure 3 depicts the DPV voltammograms of c‐Fe and c‐Zn samples obtained from the DPV analysis performed after 1, 7, and 14 days of immersion. DPV technique has been intensively used to characterize the curcumin release in a medium as well as to characterize the curcumin redox reaction based on the oxidation peaks of curcumin 32,45‐48 . In these voltammograms, two curcumin oxidation peaks can be observed after Day 7 and Say 14 of immersion for c‐Zn and after 14 days of immersion for c‐Fe.…”

“…The signal was enhanced using ECM coating before the cell culture was placed on the ECM-coated hybrid platform, indicating the ability of ECM to accelerate cell-substrate interaction. Recently, Suhito et al (2020) proposed a bio-multifunctional platform that can simultaneously perform 3D multicellular cancer spheroid formation and real-time assessment of the anticancer drug treatment (Figure 5A) [145]. The indium tin oxide (ITO) glass electrode was modified with HAuCl 4 via electrodeposition, as previously reported.…”

“…Some researchers have applied the hybrid function platform to MCTs by increasing the electrochemical signal intensity. In one study, a gold-nanostructure-based platform was designed to detect curcumin, which is known as a natural anticancer compound in a brain cancer model (Figure 7A-C) [122]. On this platform, a neuroblastoma (SH-SY5Y) and glioblastoma (U87-MG) were co-cultured to form spontaneous multicellular spheroids without any treatment.…”
