A SOX2 Reporter System Identifies Gastric Cancer Stem-Like Cells Sensitive to Monensin

Abstract: Gastric cancer remains a serious health burden with few therapeutic options. Therefore, the recognition of cancer stem cells (CSCs) as seeds of the tumorigenic process makes them a prime therapeutic target. Knowing that the transcription factors SOX2 and OCT4 promote stemness, our approach was to isolate stem-like cells in human gastric cancer cell lines using a traceable reporter system based on SOX2/OCT4 activity (SORE6-GFP). Cells transduced with the SORE6-GFP reporter system were sorted into SORE6+ and SOR… Show more

“…Oct4 high cells have more SC-like traits, such as selfrenewal, chemoresistance and xenograft tumorigenicity, than Oct4 low cells. 57 To date, Oct4 has been used to isolate CSCs by Oct4 promoter-mediated activity in breast cancer, 58 non-small cell lung cancer (NSCLC), 59 gastric cancer 60 and HCC. 61 Nanog.…”

“…Although our approach is theoretical, there is recent evidence of an invariant stem cell hierarchy in glioblastoma tumors giving rise to progenitor cells with an extensive self-maintenance capacity [22,31]. There is also evidence of subpopulations of cells showing a differential expression of embryonic stem cell markers in head and neck squamous cell carcinoma, colon adenocarcinoma, melanoma, and buccal squamous cell carcinoma [23,[33][34][35], in addition to evidence that more differentiated cells can generate tumors [9,10,17,25,28]. Moreover, recent findings on antibody-drug conjugates (ADC) showed that the targeting of Lgr5(+) colon CSCs with an ADC controlled the disease, but did not eliminate it, as observed in their reported growth curves [52].…”

“…Typically, the evaluation of stemness is assessed by the ability of sorted cell fractions to form tumors in immunodeficient mice. Despite the fact that the stem-cell-like fraction usually has an x-fold greater tumorigenic capacity compared to the non-stem-cell counterpart, the latter fraction, in some cases, also shows a reduced, but still latent, capacity to form tumors (see, for example, references [9,10,[24][25][26][27][28]).…”
