A small nucleolar RNP protein is required for pseudouridylation of eukaryotic ribosomal RNAs

Abstract: Eukaryotic rRNAs possess numerous post-transcriptionally modified nucleotides. The most abundant modifications, 2'-O-ribose methylation and pseudouridylation, occur in the nucleolus during rRNA processing. The nucleolus contains a large number of small nucleolar RNAs (snoRNAs) most of which can be classified into two distinct families defined by conserved sequence boxes and common associated proteins. The C and D box-containing snoRNAs are associated with fibrillarin, and most of them function as guide RNAs in… Show more

“…In marked contrast to Cbf5p, depletion of Gar1p does not affect the stability of the box H + ACA snoRNAs (Girard et al 1992). Gar1p depletion, however, does lead to the release of the box H + ACA snoRNA snR36 from the pre-rRNA (Bousquet-Antonelli et al 1997), indicating that it normally plays a role in establishing the snoRNA-pre-rRNA interaction or in stabilizing the base pairing.…”

“…35S pre-rRNA, 25S rRNA, and total tRNAs were purified by electroelution, digested with RNAse T2 (in 50 mM Na acetate at pH 5.5 at 37°C), and analyzed by two-dimensional cellulose TLC as described by Filipowicz and Shatkin (1983). The procedure used was essentially as described by Bousquet-Antonelli et al (1997).…”

“…This is based on the following three observations. First, all core proteins, except GAR1, are essential for the stabilization of all H/ACA RNAs (Bousquet-Antonelli et al 1997;Henras et al 1998;Lafontaine et al 1998;Watkins et al 1998;Dez et al 2001). Second, all H/ACA RNPs can be precipitated with antibodies directed against any of the core proteins, including GAR1 (Balakin et al 1996;Ganot et al 1997).…”

“…3 This result supports the idea that identical segments between the two proteins might bear the RNA binding activity. Recently, a thermosensitive allele of Gar1p has been isolated and described (29). This mutant has multiple mutations that lead to six residue changes, from which the one(s) responsible for the phenotype has not yet been mapped.…”
