A single germline VH gene segment of normal A/J mice encodes autoantibodies characteristic of systemic lupus erythematosus.

Abstract: These experiments tested the hypothesis that unmutated germline genes from normal mice can encode autoantibodies. We found that the unmutated VHIdCR gene segment, which encodes a large proportion of antiarsonate antibodies in A/J mice, also encodes antibodies with the ability to bind to DNA and cytoskeletal proteins. After Ars immunization, at a time when the VHIdCR gene segment mutates and antibody affinity for the hapten increases, reactivity with the autoantigens was lost. Six antibodies obtained after immu… Show more

“…In support of this notion is the finding that patients with systemic autoimmune diseases have increased levels of polyreactive B cells (22,23). In addition, examples exist in which NAA obtain increased autoreactivity via V gene mutation and isotype switching (24)(25)(26). In contrast, recent evidence has pointed to a role of natural IgM in maintaining selftolerance.…”

“…This hypergammaglobulinemia is often associated with autoimmunity (7, 12, 13) perhaps because of nonspecific proliferation of B cells expressing a preimmune-like repertoire that is known to be polyreactive and to recognize autoantigens (67)(68)(69)(70). Although previous spectratypic analyses show that B cell HCDR3s from PRRSVinfected piglets were a signature for polyclonal activation, certain HCDR3 lengths were pronounced (12).…”

“…Such a progression was observed in use of the unmutated 36 -65 V H gene segment in IgM ssDNA-binding natural autoantibodies before immunization and in IgM and IgG Abs to the Ars hapten after immunization (20). In that case, however, it was not possible to follow the fate of a particular rearranged V H D H J H combination.…”

“…A significant fraction of hybridomas made from LPS-stimulated lymphocytes of normal newborn mice (17) or nonimmunized adult mice (18,19) makes polyreactive IgM autoantibodies that bind ssDNA and have unmutated V domains (20). Anti-DNA Abs are also produced by normal B lymphocytes activated by EBV in tissue culture (21).…”
