Gangliosides are known to be developmentally regulated and regionally variable, but these variations have not been shown to occur among precisely defined nuclei of the brain in relation to either aging or function. We have sought to correlate changes in ganglioside distribution with age-related changes in highly specific brain regions known to control a common function, the regulation of rapid eye movement sleep architecture. Gangliosides were extracted and quantified from micropunched regions of the locus coeruleus, dorsal raphe, laterodorsal tegmentum, pedunculopontine tegmentum and the general region of the pons containing these nuclei in young adult (3 months), adult (12 months), and aged (24 months) rats. The ganglioside distribution patterns were generally characteristic of the pons as a whole, but showed a high level of differentiation in time course at specific anatomical sites.

Ganglioside content and pattern were analyzed in the rat hippocampus and in the retina and optic tectum of the chick embryo during periods of maximum neural differentiation. Allometric plots of ganglioside sialic acid against tissue dry weight were linear on a log-log scale, in either one (hippocampus and retina) or two (optic tectum) phases and at unique rates for each tissue. Ganglioside patterns changed consistently during development with decreases in D3 and increases in M1 and D1a. The ratio of D3 to M1 + D1a was found to be a simple quantitative predictor of the extent of differentiation (particularly synaptogenesis) in each tissue. However, a full complement of gangliosides was found prior to the onset of significant synapse formation.

Cholinergic murine neuroblastoma cells, maintained in vitro, were exposed to a low concentration (0.4 μg/ml) of adriamycin. Morphologically the treated cells appeared to differentiate. The cell bodies increased in size from an average fixed cell body diameter of 7–13 to 25–40 μm, the cells developed long processes, became argyrophilic and the percentage of cells undergoing mitosis decreased relative to controls. Acetylcholine esterase activity increased in the drug-treated cells suggesting induction of differentiation. However, choline acetyltransferase activity and ganglioside composition remained unchanged. In addition, inoculation of mice with 2 × 10⁵ viable drug-treated or control cells resulted in all of the mice developing neuroblastoma. No differences were observed in either the rate of tumor development or survival times. These results suggest that neuroblastoma cells may survive adriamycin treatment by becoming ‘differentiated’, ceasing cell division until conditions favor their undergoing another cell cycle.
