A Simple Method for the Design and Development of Flavivirus NS1 Recombinant Proteins Using an In Silico Approach

Abstract: Even in countries that are currently not facing a flavivirus epidemic, the spread of mosquito-borne flaviviruses presents an increasing public threat, owing to climate change, international travel, and other factors. Many of these countries lack the resources (viral strains, clinical specimens, etc.) needed for the research that could help cope with the threat imposed by flaviviruses, and therefore, an alternative approach is needed. Using an in silico approach to global databases, we aimed to design and devel… Show more

“…The UniprotKB accession numbers for aligned sequences are as follow: DENV-1, P27909; DENV-2, P29991; DENV-3, Q6YMS3; DENV-4, Q2YHF0; JEV, P27395; TBEV, Q01299; WNVm, P06935; YFV, Q6J3P1; and ZIKV, Q32ZE1. The conserved regions are indicated in red, whereas the non-conserved regions are in white . The conservation scale shown in yellow refers to the degree of similarity between amino acid or nucleotide residues at a particular position in a sequence across different members of a group or family of related sequences.…”

“…The conserved regions are indicated in red, whereas the non-conserved regions are in white. 94 The conservation scale shown in yellow refers to the degree of similarity between amino acid or nucleotide residues at a particular position in a sequence across different members of a group or family of related sequences. polyprotein is directed to the ER because of the final 24 amino acids in the E protein, which functions as a signal sequence, translocating the NS1 protein into the ER lumen.…”

“…Recombinant NS5 was produced in HEK293T cells for phosphorylation characterization [ 73 , 74 ]. Recombinant YFV NS1 was obtained in E. coli strain Lemo21 (DE3) using vector pBT7-N-His [ 75 ]. NS1 and E proteins were also produced in insect and mammalian cells [ 76 ].…”
