A Simple Method for Human Whole Blood Microcultures and Its Application in Radiation Biodosimetry

Abstract: The scoring of the cytokinesis-block micronucleus and dicentric chromosomes in human peripheral blood lymphocytes is used as a dosimeter of radiation exposure. A detailed methodology is presented for human whole blood microculture for cytogenetic analysis. The technique described yields more than sufficient numbers of mitotic lymphocytes for analyzing micronuclei and chromosome aberrations following exposure to radiation.

“…At the same time, this method can make us from the traditional manual recognition to the automation of recognition. 7,13,14,15,16 When preparing dose-response curves, Should as far as possible meets the demand of statistics, analysis of cell number formula is based on a chromosome aberration rate in line with the binomial distribution, if known aberration cell rate and the permissible error, can according to the binomial distribution calculated 95% confidence limit formula should count the number of cells, biological experimental error tolerance for general use 15%, it need to count a considerable number of cells, currently use 20% more The formula is: N = [(1 − p) × 96.04]/p. To improve the confidence limit of the results, a sufficient number of cells should be analyzed, and the number of cells to be analyzed depends on the distortion rate.…”

“…The SARRP uses a radiation therapy planning (13), verification (14) and delivery system called MuriPlan that allows for both planning and delivering radiation therapy. The MuriPlan planningmodule enables single or multiple beams to be targeted to the isocenter and can create contours of tumors and of organs at risk to reduce dose to normal tissues.…”
