A Simple Method for Determination of Protodioscin in Tribulus Terrestris L. and Pharmaceuticals by High-Performance Liquid Chromatography Using Diode-Array Detection

Abstract: Abstract:The Tribulus terrestris L. (Zygophyllaceae) is a known medical plant used in traditional and folk medicine worldwide. The steroid saponine protodioscin, an active component found in this plant, serves as a marker for quality control of plant raw materials. In this study, we developed a simple and selective method for determination of protodioscin using convenient High-Performance Liquid Chromatographic (HPLC) laboratory equipment. The detection was performed by Diode-Array Detector (DAD). The proposed… Show more

“…Researchers have identified several components in the TT extract, especially steroidal saponins, flavonoids, tannins, terpenoids, polyphenol carboxylic acids, and alkaloids [98]. Protodioscin [Figure 5], as the main active component of steroidal saponins, is thought to have a critical role in the pharmacological process of TT [99,100].…”

“…Some studies have shown that steroidal saponin and cinnamic acid amide compounds are responsible for the antidiabetic, anti-inflammatory and antioxidant activities of T. terrestris extracts (Ernawati et al, 2017;Kim et al, 2018;Lee et al, 2017;Zhu et al, 2017). According to several studies, protodioscins are the main components of the aerial parts of T. terrestris (De Combarieu et al, 2003;Kostova et al, 2002;Shishovska et al, 2015). Although saponins show weak UV absorption, even in the short wavelength range, Shishovska et al (2015) reported that the HPLC-DAD method is suitable for routine control of raw plant material and pharmaceuticals based on T. terrestris.…”

“…According to several studies, protodioscins are the main components of the aerial parts of T. terrestris (De Combarieu et al, 2003;Kostova et al, 2002;Shishovska et al, 2015). Although saponins show weak UV absorption, even in the short wavelength range, Shishovska et al (2015) reported that the HPLC-DAD method is suitable for routine control of raw plant material and pharmaceuticals based on T. terrestris. To investigate the presence of protodioscin in the samples, the protodioscin peak from the sample chromatograms was identified by the retention time by comparison with the protodioscin standard chromatogram, co-injection with a reference compound and UV210 spectrum matching.…”

“…The plant grows in warm and tropical areas of Europe, Asia, Africa and Australia. In Ukraine, caltrop is encountered in the southern regions (Raina et al 2007;Mohd et al 2012;Chhatre et al 2014;Shishovska et al 2015).…”

“…The caltrop herb contains the steroid saponins: protodioscine, dioscine, prototribestin, pseudoprotodioscin, tribestin, tribulosin, gracillin, protogracillin, diosgenin, gitogenin, ruscogenin, gecogenin, tigogenin, chlorogenin and sarsasapogenin (Dinchev et al 2008;Mohd et al 2012;Hammoda et al 2013;Kang et al 2014;Shishovska et al 2015). Besides them, its components are also flavonoids, tannins and nitrogen-containing substances (Sidjimova et al 2011;Kumar and Bhardwaj 2012).…”
