Culex ßavivirus (CxFV) is an insect-speciÞc ßavivirus that was Þrst reported in 2007 in Japan. CxFV strains were isolated from Culex tritaeniorhynchus Giles and Culex pipiens L. group mosquitoes and genetically characterized in Toyama Prefecture, Japan, from 2004 to 2009, to reveal host speciÞcity, mode of transmission, and seasonal and geographical distribution. The minimum infection rate (MIR) of CxFV within Cx. tritaeniorhynchus populations was 0.3 and much lower than that within Cx. pipiens group (17.9). The complete genome sequences of 11 CxFV isolates (four from Cx. tritaeniorhynchus and seven from Cx. pipiens group) consisted of 10,835Ð10,837 nucleotides. When these 11 isolates and Þve reference strains (NIID-21-2 and Tokyo strains from Japan, Iowa07 and HOU24518 strains from the United States, H0901 strain from China) were compared, there were 95.2Ð99.2% nucleotide and 98.1Ð99.8% amino acid identities. Phylogenetic analysis showed that the 11 isolates were divided into four clusters. One cluster consisted of Þve isolates from Cx. pipiens group and Cx. tritaeniorhynchus from one site and their nucleotide sequences almost completely matched. One cluster consisted of an isolate with a unique sequence from a Cx. pipiens group mosquito captured in an aircraft from Taiwan, suggesting that it was introduced from abroad. CxFV strains were divided into several groups according to countries when nucleotide sequences of CxFV available in GenBank and 11 Toyama isolates were compared. These results suggest that CxFV is maintained in nature among Culex mosquitoes in a mosquito habitat-speciÞc but not a species-speciÞc manner.

Mosquitoes were collected weekly from May 2003 to December 2013 at a pair of sites within the premises of the National Institute of Infectious Diseases, Tokyo, Japan. The mosquito trap was equipped with 1 kg dry ice and battery-operated suction mechanism. A total of 10,546 adults belonging to 7 species in 3 genera were collected in this study. Aedes albopictus and Cx. pipiens gr. were the dominant mosquitoes, composing 99.7% of the total mosquitoes collected. Six and 12 adults of Cx. tritaeniorhynchus were collected in early spring and in late summer to autumn, respectively. More Cx. pipiens gr. were collected at the high trap site near a tree canopy rather than the low trap site near the ground every year, whereas only 1 to 15% of Ae. albopictus were collected at the high trap site, indicating a difference in searching behavior between the two species. The female density of Cx. pipiens gr. was high (>10/trap/day) during early June to early August, and a high Cx. pipiens gr.-related risk is expected in this period. Density of female Ae. albopictus was high (>10/trap/day) during the 28th to 41st week, suggesting a high risk of transmission of Ae. albopictus-borne pathogens from mid-July to mid-October in Tokyo.

To investigate the possible spread of West Nile virus (WNV) into Japan, we carried out entomological surveillance for flaviviruses at migratory bird stopover sites in Hokkaido, Japan, during 2003-2006. A total of 3,826 mosquitoes, identified as 15 species in five genera, were collected and 2,465 of these were grouped into 123 pools that were assayed for cytopathic effects on mosquito and mammalian cell cultures and for flavivirus RNA by reverse transcription-polymerase chain reaction using flavivirus universal primer sets for fragments of the NS3 and NS5 genes. Neither WNV nor other mosquito-vertebrate transmitted flaviviruses were detected in mosquitoes collected at any of the sites in Hokkaido, but five Culex flaviviruses and one novel Aedes galloisi flavivirus were identified from Culex pipiens L. s. l. and Aedes galloisi Yamada, respectively. Genetic and phylogenetic analyses based on the partial NS5 nucleotide sequences classified Aedes galloisi flavivirus with the insect flavivirus, but distant from Cell fusing agent, Kamiti river virus, and Culex flaviviruses, showing <74% sequence identities. Polymerase chain reaction-based bloodmeal analysis of 79 females showed that all of the Aedes and Ochlerotatus mosquitoes fed on mammals (deer and humans), whereas, Cx. pipiens s. l. mosquitoes fed on both of avian (ducks and sparrows, 85.7%) and mammalian hosts (dog, 14.3%). We suggest that to date WNV has not become established in Japan.
