The expensive beef have encouraged counterfeiting beef on processed food products such as meatballs. Mice meat is frequently reported used for adulteration of beef. The accurate method is needed to ensure the supervision of food safety. This study reports the use of DNA testing to detect the presence of mice meat in meatballs with real-time PCR primer specific. PCR primers designed based on the ND-1 gene of mice mitochondrial DNA with the sequence are 5’-CGGCATCCTACAACCATTTGC-3’ and 5’-CGGCTCGTAAAGCTCCGAA-3’, respectively, target 294 bp DNA fragment. The real-time PCR can specifically detect the presence of the mice meat in a meatball with no detection the presence of beef, mutton, chicken, pork, and horsemeat. The method showed good precision shown by the CV of repeatability test at 2%, much lower than the requirement of < 25%. Real-time PCR was able to deliver positive results for as low as 0.5 ng DNA template, equivalent to 0.08 copies of genome DNA of mice equal to 80–150 copies of mtDNA. By using standard phenol-chloroform DNA isolation technique, this method is able to detect contamination of mice meat in meatball up to 1%. Three commercial meatballs confirmed contaminated by mice meat using the method.

Background: Food fraud is described as a violation of food law, which is intentionally committed to get an economic or financial gain through the consumer's swindle resulting in multi-million business and posing a public health threat. The main fraudulent practices are mislabelling of composition, certificates of origin, health claims, and artificial increases in weight or volume caused by replacement, dilution, addition or removal of some ingredients. Hardly 68% of the food fraud violations are produced in animal and vegetable products with high fat content (27% meat, 13% fish, 11% oils, 10% dairy products, 4% nuts and seeds and 3% animal by-products) becoming a crucial issue for food processing industries. Scope and approach: The present review focuses on the main authentication techniques and methods employed to clarify the authenticity of both animal and vegetable fat food products emphasizing the importance of the use of robust and reliable analytical techniques combined with multivariate analyses. Key findings and conclusions: Targeted approaches, such as as chromatography and DNA-based methods, combined with multivariate analysis have shown high accuracy, sensitivity and selectivity, allowing the simultaneous evaluation of multiple analytes. In addition, non-target methods, such as those based on spectroscopic techniques, have been used to establish the geographic origin of food products with quick response, low cost, non-destructive character and also offering the possibility to be miniaturized.

A multiplex PCR assay was developed to simultaneously differentiate four antelope species and identify adulteration in Cornu Saigae Tataricae. Four novel primer sets were designed with high inter-species specificity and intra-species stability. Limit of detection was estimated to be 10 ng of genomes. When a mixture of antelope hornand fake species was assayed, it exhibited powerful differentiation capability. 5 out of 12 batches of commercialproducts were identified to be counterfeited or adulterated with Ovis aries Linnaeus and/or Capra hircus Linnaeus. It is readily applicable in routine analysis for identification of sham or adulterants of Cornu Saigae Tataricae.
