A simple and sensitive colorimetric pH meter based on DNA conformational switch and gold nanoparticle aggregation

Abstract: A simple and rapid colorimetric pH meter has been developed based on the conformational switch of i-motif DNA and non-crosslinking AuNP aggregation, the average accuracy of the nano-meter was found to be +/-0.04 pH unit across the physiological operating range.

“…The degree of Au NP aggregation is expected to be influenced by the PCR products which serve as the linker molecules for forming the Au NP agglomeration. More cycles of the PCR procedure will lead to more PCR amplification which provides more linker molecules, and will result in a larger red-shift of UV-visible spectrum [21]. As expected, the spectral shift increased when the cycle number N was increased from 5 to 20 in our experiments (Fig.…”

“…The presence of NaCl could induce the formation of AuCl 4 À ions, which would lead to slow reaction kinetics and the subsequent formation of uniform shell structures (Fig. [35][36][37] The strong interactions between the adenine base and the gold surface are predominant in acidic pH conditions. “…477 Gold-functionalised nanoparticles are more widely used, and are useful as they undergo a colour change from red to purple on aggregation, as well as possessing surface plasmon properties. The former has been used to detect hybridisation, 478,479 to detect conformational switching in i-motif sequences, 480 for the detection of mismatches using the repair enzyme MutS, 481 and linear amplification of DNA using Bst DNA polymerase. 482 Gold-conjugated oligonucleotides have been used to prepare crystalline assemblies using the complementarity of nucleotides, which may be of use in nanodevices.…”
