A simple and cost-effective approach to fabricate tunable length polymeric microneedle patches for controllable transdermal drug delivery

Abstract: Polymeric microneedles (MNs) are attractive transdermal drug delivery systems because of their efficient drug delivery and minimal invasiveness. Master template fabrication is the most time-consuming and costly step in producing polymeric MNs using a micromoulding approach. Herein, this issue is addressed by modifying tattoo needle cartridges by adjusting the volume of a PDMS spacer, thus streamlining polymeric MN fabrication and significantly reducing its manufacturing cost. Using the fabricated master templa… Show more

“…Polymeric MN-based natural polysaccharide is less expensive than silicon MNs or metal MNs [155]. Polymers such as chitosan, chitin, hyaluronic acid, sodium alginate and starch are used for the fabrication of microneedles due to their low cost [155].…”

Polymers such as chitosan, chitin, hyaluronic acid, sodium alginate and starch are used for the fabrication of microneedles due to their low cost [155]. Furthermore, the current microneedle manufacturing processes need to be enhanced to achieve largescale production to completely transfer microchip-based microneedles into therapeutic applications.…”

“…Use of heat or UV light during the procedure can critically affect the loaded drugs. However, it is a simple, reproducible and scalable process and some improvements have been recently reported ( Chen Y. et al, 2020 ). Drawing lithography is a direct patterning process based on viscosity and elastic deformation of polymers in glass transitions, thereby avoiding the need of mask and light irradiation but requiring polymer curing and with limitations when biodegradable materials are intended to be used ( Lee and Jung, 2012 ).…”

“…The experience gained from the early MEMS applications has tailored their emerging technology for biotechnological developments including water and environmental monitoring, DNA sequencing and drug discovery ( Bashir, 2004 ). Novel drug delivery methods to release a specific dose of a drug with a certain rate, during an adequate period of time in the targeted tissue and facilitating patient self-administration are becoming increasingly necessary ( Orive et al, 2003 ; Geipel et al, 2007 ; Ruggiero et al, 2018 ; Singh et al, 2019 ; Tharkar et al, 2019 ; Chen Y. et al, 2020 ; Deng et al, 2020 ; Hwang et al, 2020 ). In this sense, advanced biomedical technology will comprise implantable MEMS devices of proven biocompatibility ( Voskerician et al, 2003 ), to carefully release drugs into the body from micro-chambers hosted in the device, eliminating the need for injections or needles ( Grayson et al, 2004 ; Vasudev and Bhansali, 2013 ).…”

“…Bee venom delivering dosages must be conducted repeatedly to enhance the disordered condition over a planned period. Therefore, it may help to develop means to assist patients to take the medication without the hassle of performing self-injections (when the presence of a medical personal is not available) (Schiff et al, 2017 ; Jung & Jin, 2021 ), also produce an economical administrative solution (B. Y. Lee et al, 2015 ; Chen et al, 2020 ) that causes minimal pain. MN delivery system is an emerging area for both the systemic and local delivery of macromolecules and helps generalize the application of medicine (Bariya et al, 2011 ; Tran et al, 2021 ; Zhang et al, 2021 ).…”
