A shift from papillary to reticular fibroblasts enables tumour–stroma interaction and invasion

Abstract: Conversely, our results show that Pfs contain cancer cells more within the epidermis. Rfs are clearly predisposed to differentiate into CAFs upon SCC signals, assisting invasion and EMT.

“…Epidermis and dermis were separated after removal of adipose tissue followed by enzymatic digestion and primary cell suspensions were obtained and cultured as described before [3,55]. Isolated primary keratinocytes and fibroblasts were tested for mycoplasma contamination using polymerase chain reaction (PCR) analysis before further use.…”

“…Sophisticated in vitro tools which resemble native human skin (NHS) to a high extent are three-dimensional human skin equivalents (HSEs). Therefore, these are widely applied in preclinical screenings and for research purposes to increase understanding of skin biology and epidermal barrier formation in healthy and diseased phenotypes [1,2,3,4,5,6]. HSEs mimic NHS in morphologic appearance, including the presence of distinguishable epidermal layers and formation of the SC [7,8,9].…”

“…the complement activation pathway (Stunova and Vistejnova, 2018). On the other hand, high expression of transglutaminase 2, calponin1, cadherin2, and matrix gla protein in Frs indicates that Frs express genes involved in cytoskeleton dynamics and cell motility (Shinde et al, 2017;Hogervorst et al, 2018). However, there also seem to be markers, such as CD26, that change dynamically with external factors.…”

“…To date there have been few studies of how different fibroblast lineages contribute to tumour stroma formation, and whether the tumour stroma differs between different types of skin cancers. Interestingly, one study showed that fibroblasts of the reticular dermis are predisposed to differentiate into CAFs upon cSCC signals, assisting invasion and EMT [60].…”
