A sequential protocol combining dual neuroanatomical tract-tracing with the visualization of local circuit neurons within the striatum

Abstract: We describe here an experimental approach designed to aid in the identification of complex brain circuits within the rat corpus striatum. Our aim was to characterize in a single section (i) striatal thalamic afferents, (ii) striatopallidal projection neurons and (iii) striatal local circuit interneurons. To this end, we have combined anterograde tracing using biotinylated dextran amine and retrograde neuroanatomical tracing with Fluoro-Gold. This dual tracing protocol was further implemented with the visualiza… Show more

“…The latter is usually done via an antibody that highlights a specific type of neuron (presumed GABAergic, peptidergic, cholinergic, or dopaminergic targeted neurons, to name just a few). Initial attempts in this realm included experiments with 'absorption' tags, that is visualization with different light absorption ('colored') chromogens such as diaminobenzidine (brown precipitate), nickel-enhanced diaminobenzidine (black precipitate), tetramethylbenzidine (dark blue precipitate), and Vector VIP™ (magenta precipitate) (Wouterlood et al 1987;Lanciego et al 1997Lanciego et al , 2000Köbbert et al 2000;Gonzalo et al 2001;Lanciego and Wouterlood 2011 (review); Unal et al 2014), while the introduction of confocal laser microscopy, and in parallel the introduction of robust fluorochromes, delivered resolution high enough to reliably detect markers even inside thin fragile fibers and axon varicosities (Wouterlood et al 2007). Most work, nowadays, published in this field is documented with confocal microscopy (e.g., Boulland et al 2009;Barbier et al 2018).…”

“…The present report is focused on the analysis of thalamostriatal and thalamo-subthalamic circuits arising from the PF nucleus by using multiple neuroanatomical tracing methods (Gonzalo et al, 2001). Emphasis was placed on the elucidation of the presumptive postsynaptically targeted neurons within both the striatum and STN.…”

“…Nickel salts have been used at much higher concentrations in DAB–Tris than any other metal (Hancock, 1982, 1984; Tago et al , 1986; Lanciego et al , 1997; Tajima et al , 2000; Gonzalo et al , 2001). To further investigate this phenomenon, dot blots were prepared, as described earlier, from Tris‐buffered DAB solutions containing 1, 5, 20 or 100 mM NiCl 2 and incubated, with or without treatment with 0.3% Na 2 S, for 10 min in developer.…”
