A Sensitive Small-Volume UV/Vis Flow Cell and Total Absorbance Detection System for Micro-HPLC

Abstract: An optical multichannel absorbance detection system suitable for micro-HPLC is described. The detection system includes a flow cell with detection volume of 30 nL and path length of 12 mm. A fiber optic spectrometer with charge-coupled device array is used to collect spectral information at 1 Hz frequency. Signal-to-noise ratios are enhanced through the use of large bandwidth total absorbance signals, while linearity and spectral resolution are maintained. Theoretical predictions of bandwidth dependent signal-… Show more

“…[1][2][3][4] As an example, smaller sample volumes allow reduced dead volumes in flowing systems and thereby limit the spread of bands flowing through a detection cell. 5 We observed enhanced detection in a previous series of studies [6][7][8] of the flow of supercritical fluids in microcapillary columns (250 mm i.d.) packed with 5 mm diameter silica particles.…”

“…On-line coupling of the miniaturized sample preparation technique and the microcolumn separations allow the hyphenated system to be further miniaturized; indicating the future possibility of developing a microscale total analytical system (micro-TAS) [131 -139], although the down-sizing of the detection device [140,141] should be additionally accomplished.…”

“…acrolein, glutaraldehyde, and dialdehydes) based on the cartridge method has encountered stability problems with the DNPH hydrazones [11]. Modifications in analytical procedure in terms of the type of adsorbents, the derivatization reagent, as well as the utilization of other analytical measurement principles have been reported in recent years [1,3,4,[13][14][15][16][17]. Owing to interference, ozone has to be removed by employing a KI denuder or scrubber upstream to the carbonyl collection.…”
