A Sensitive and Effective Proteomic Approach to Identify She-Donkey’s and Goat’s Milk Adulterations by MALDI-TOF MS Fingerprinting

Abstract: She-donkey’s milk (DM) and goat’s milk (GM) are commonly used in newborn and infant feeding because they are less allergenic than other milk types. It is, therefore, mandatory to avoid adulteration and contamination by other milk allergens, developing fast and efficient analytical methods to assess the authenticity of these precious nutrients. In this experimental work, a sensitive and robust matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) profiling was designed to a… Show more

“…The proteomics method as an emerging method with high sensitivity and selectivity has also been applied in the determination of milk adulteration. This method focuses on qualitative (Di Girolamo et al, 2014;Bernardi et al, 2015) or semiquantitative (Camerini et al, 2016) analysis. Milk adulteration driven by economic reasons affects the percentage of milk in dairy products.…”

“…Among other applications, it has been used to determine the origin of food [69][70][71] and food adulterations [72][73][74][75][76][77]. The discrimination of samples on the basis of their protein fingerprints using MALDI-MS is also an intense area of research [78][79][80].…”

“…Similar limits of adulteration (ca. 0.5 %) were also obtained through MALDI-TOF MS making it possible to detect adulteration of both donkey and goat milks with other species' milks (cow, ewe, and buffalo) on the basis of their different proteomic profiles [77].…”

“…To detect adulteration of goat's milk with cow's milk the reference method is used, which is based on the differentiation of isoelectric points of cow and goat γ 2 -and γ 3 -caseins originating from the hydrolysis of the belonging β-caseins (Commision Regulation 2008; EC 273/08). But there are many studies, where mass spectrometry (MS) (C hen et al, 2004; C alvano et al, 2012; Girolamo et al, 2014) or gel based approaches (Hinz et al, 2012;Špoljarić et al, 2013;Yang et al, 2014) have been used to detect adulteration of goat's milk with cow's milk or just for comparing protein profiles of different animal species milks. Although the general tendency nowadays is to move to gel-free workflows, protein electrophoresis has been used extensively (Ortea et al, 2012) and continues to be used (mainly 2-DE) in food authentication studies (Ortea et al, 2016).…”
