A semi-selective medium for detection, isolation and enumeration ofErwinia carotovora ssp.carotovora from plant materials and soil

Abstract: Using a modified crystal violet sodium polypectate (CVPB) double layer medium with 2,3,5-triphenyltetrazolium chloride and L-asparagine in the basal medium and bromthymol blue in the upper, both containing tryptone, gave red to pink colonies of Erwinia carotovora ssp. carotovora on a blue green background with cavity formation. The addition of 1.2 µg/ml polymyxin B sulphate to the upper layer inhibited the growth of Erwinia carotovora ssp. atroseptica, while 32 µg/ml erythromycin in the upper layer inhibited t… Show more

“…In particular, double‐layer modifications have recently been used to isolate species of both Pectobacterium and Dickeya (Hyman et al. , 2001; Bdliya & Langerfeld, 2005). Incubation on this medium with differential temperatures and erythromycin susceptibility has been proposed for selective isolation of P. atrosepticum , P. carotovorum and Dickeya spp.…”

“…Cada muestra se inoculó en tres tubos de ensayo que contenían agua estéril, caldo nutritivo y medio líquido de pectina (polipectato de sodio al 75 %, de (NH 4 ) 2 SO 4 al 10 %, de Mg 2 SO 4 7H 2 O al 5 %, y de K 2 HPO 4 al 10 %) (Bdliya & Langerfeld, 2005). Los tubos con agua se agitaron durante cinco minutos a 50 r.p.m.…”

“…There are several methods used to identify and detect Dickeya species, and D. dianthicola specifically. The selective medium CVP is commonly used to detect Pectobacterium and Dickeya isolates ( Cuppels and Kelman, 1974;Hyman et al, 2001; Bdliya and Langerfeld, 2005;Hélias et al, 2012). In general, Dickeya isolates can mostly be differentiated from Pectobacterium isolates based on differences in growth temperature, i.e., growth above 36 C selects for Dickeya species (Pérombelon and Hyman, 1986).…”
