A semi-automated neutral red based chemosensitivity assay for drug screening

Abstract: A semi-automated colorimetric chemosensitivity assay was developed. The assay utilizes the vital stain neutral red for the rapid screening of potential anticancer agents using solid tumor cell lines. The cell lines used in this assay and presented in this report are CX-1 colon adenocarcinoma and A549 lung carcinoma. The assay, performed in 96 well tissue culture plates, allows for short drug exposure times (3 hrs.) followed by quantitation of cell number (neutral red absorbance) following four cell doubling ti… Show more

“…The usual correlation between visual and NR assays in our hands has been greater than 95% (Barnard et al, 2001). The NR assay was performed using a modified method of Cavenaugh et al (1990) as described by Barnard et al (1999).…”

“…To name a few specific examples, the determination of daughter fates before the division of neural progenitor cells (22), the existence of haemogenic endothelium (23), neural and hematopoietic stem and progenitor divisional patterns and lineage choice (24,25), the in vitro tissue culture studies using the neutral red dye (26), the studies of dynamics of collective cell migration (27), detection of toxic compound (28), and drug screening (29,30). In these cases, the labor-intensive nature of these experiments and the challenge of efficiently imaging large assays have typically plagued this type of experiment format.…”
