A self‐contained polymeric 2‐DE chip system for rapid and easy analysis

Abstract: We developed a polymeric 2-DE chip system. The chip consisted of an IEF region, an SDS-PAGE region, a valveless connection port, and a sample introduction port. A "junction structure" as a valveless connection port, which allowed separating and connecting the first- and second-dimensional gels, was fabricated between their regions. A "solution inlet" as a sample introduction port was fabricated to perform the liquid and sample introductions without solution leakage. Simultaneous sample monitoring was performed… Show more

“…Due to the aforementioned advantages of thermoplastics, efforts have been made in implementing 2DGE in thermoplastic devices. 26,53,[77][78][79][80][81][82][83] Table 2 lists these efforts in the chronological order of the publications. An early example by Griebel et al was a credit-card-sized PMMA device, which was comprised of an IEF slot in the top layer and 300 parallel PAGE channels in the bottom layer (Fig.…”

“…Fluorescence imagers have also been used as detectors for SDS-CGE [19, 90, 114, 115]. A fluorescence imager is a great tool for early stage technology development since it allows researchers to see the migration of proteins inside a capillary or a microfabricated channel.…”

“…To meet the requirement, a barrier or mechanism is required to achieve introduction of different separation media into two dimensions and isolation of two media at the same time. In the work discussed above, a variety of methods were used for the interface, including vertical through-holes in multiple layers [9], viscous media [45], smaller-channel connections [48], off-line transport of an IPG strip [49], and a connection port [50]. However, these mechanisms are either too complicated, or imperfect, or difficult to automate.…”

“…Transfer from the first to the second dimension was demonstrated, though no result was reported for protein separation. Yokoyama and co-workers employed an IPG strip for IEF, followed by SDS-PAGE in a PMMA device with meso-scale features [50,51]. The setup was very similar to conventional 2-DE, except that it was miniaturized.…”
