A self-assembling peptide reduces glial scarring, attenuates post-traumatic inflammation and promotes neurological recovery following spinal cord injury

Abstract: The pathophysiology of spinal cord injury (SCI) involves post-traumatic inflammation and glial scarring which interfere with repair and recovery. Self-assembling peptides (SAPs) are molecules designed for tissue engineering. Here, we tested the performance of K2(QL)6K2 (QL6), a SAP that attenuates inflammation and glial scarring, and facilitates functional recovery. We injected QL6 into the spinal cord tissue of rats 24 h after clip compression SCI. QL6 led to a significant reduction in post-traumatic apoptosi… Show more

“…QL6, one of these self-assembled nanomaterial peptides, preserves spinal cord tissue upon injection into rats 24 h after spinal cord injury. The mode of action of QL6 is suggested to be a significant reduction in apoptosis, inflammation, and astrogliosis [53].…”

“…Liu et al used peptide QL6, which self-assembles into-sheets at neutral pH, as their therapeutic agent for SCI [15]. This study found a favorable distribution of the nano bers after injection and almost full degradation by eight weeks.…”

“…Although QL6 nanofibers themselves might improve cell survival 4,15 , nevertheless, the use of growth factors seems to be advantageous 16,17,18,19,20,21 . They can be administered either by hydrogels releasing growth factors 22 , or by application via osmotic pumps (as described above).…”

“…In order to provide good tissue conditions for NPC survival and integration, several studies have chosen injection points in the adjacent white matter, 2 mm rostral or caudal to the lesion site and not directly in the epicenter of the lesion 4,15,18 . There, NPCs have a better chance for survival, integration, and differentiation into astrocytes, oligodendrocytes or neurons, and can migrate towards or into the lesion resulting in axonal sprouting and improved axonal connectivity.…”
