A Selective Activity-Dependent Requirement for Dynamin 1 in Synaptic Vesicle Endocytosis

Abstract: Dynamin 1 is a neuron-specific guanosine triphosphatase thought to be critically required for the fission reaction of synaptic vesicle endocytosis. Unexpectedly, mice lacking dynamin 1 were able to form functional synapses, even though their postnatal viability was limited. However, during spontaneous network activity, branched, tubular plasma membrane invaginations accumulated, capped by clathrin-coated pits, in synapses of dynamin 1-knockout mice. Synaptic vesicle endocytosis was severely impaired during str… Show more

“…2) and dynamin-catalyzed fission 58,67 . In agreement with this, dynamin 1 knockout mice exhibit a striking, activity-dependent accumulation of tubular coated endocytic intermediates 48 . Data from dynamin temperature-sensitive mutant flies (shibire ts ) suggest a possible role for dynamin that may go beyond its established role in endocytic membrane fission (see below).…”

“…Molecular analysis using genetic, biochemical and optical imaging approaches have provided strong evidence for an important, if not essential, role of clathrin-and dynamin-mediated endocytosis in synaptic vesicle reformation 11,48 . Acute chemical, sustained genetic or RnAi-mediated perturbation of clathrin 11 or dynamin function in a variety of organisms and model systems 7,48-51 causes strong defects in synaptic vesicle recycling.…”

“…Block of dynamin I function has no effect on the final endosomal scission step under elevated stimulation conditions where a dynamin II and clathrin mediated process predominates in adrenal chromaffin cells [4,22]. This seems not to extend to neuronal preparations where dynamin I seems to be directly associated with clathrin-mediated internalization of small, clear vesicle membrane [46][47][48] where it is activated under elevated stimulus conditions [26,[49][50][51]. This point is of note for several reasons.…”

“…Chronic disruption of dynamin I may result in overall poor cell health. Genetic knock out of dynamin I results in compromised animal health and viability [26]. In order to guard against non-specific effects of chronic dynamin I disruption, we chose an alternate disruption approach.…”
