A Secreted BMP Antagonist, Cer1, Fine Tunes the Spatial Organization of the Ureteric Bud Tree during Mouse Kidney Development

Abstract: The epithelial ureteric bud is critical for mammalian kidney development as it generates the ureter and the collecting duct system that induces nephrogenesis in dicrete locations in the kidney mesenchyme during its emergence. We show that a secreted Bmp antagonist Cerberus homologue (Cer1) fine tunes the organization of the ureteric tree during organogenesis in the mouse embryo. Both enhanced ureteric expression of Cer1 and Cer1 knock out enlarge kidney size, and these changes are associated with an altered th… Show more

“…Whole E15.5 kidneys were fixed in 4% paraformaldehyde (PFA) and processed for OPT as described (Chi et al, 2011) with some modifications. Fixed kidneys in TBST (Tris-buffered saline with 0.01% Triton X-100 and 10% foetal bovine serum) were stained with anti-nephrin antibodies (a kind gift of K. Tryggvason, Karolinska Institute, Stockholm, Sweden) at 4°C for 3 days and then extensively washed in TBST.…”

“…Regulation of BMP ligands by extracellular antagonists is a critical mechanism for numerous developmental programs, including germ layer specification and temporospatial gradients important for the establishment of the dorsal-ventral axis and organ and skeletal formation 7; 8; 9; 10; 11; 12; 13 . In addition to their role in development, BMP antagonists also have roles in the adult.…”

“…Encountering of cells within an individual niche appears to be adjusted by the secreted bone morphogenetic protein antagonist Cerberus homolog1, Ret proto oncogene, and ETS translocation variant4 [59, 60]. The molecular motion triggers the tip of a CD ampulla in precise position to the nearest cells of mesenchyme.…”
