A second capsidless hadakavirus strain with 10 positive-sense single-stranded RNA genomic segments from Fusarium nygamai

Abstract: Sequence data availability. The HadV1-1NL complete nucleotide genomic sequence was deposited in the GenBank/ENA/DDBJ database under accession numbers LC592214 to LC592223.

“…NGS sequencing provided many virus-like sequence contigs. Because the NGS data were from three fungal strains (see Materials and Methods), sequence contigs associated with previously reported mycoviruses derived from two other fungi ( Khan et al., 2021a ; Khan et al., 2021b ) were eliminated from the current analyses. RT-PCR analyses assigned a total of 17 virus-like sequence contigs to strain L3.…”

“…The fungal strain L3 ( D. seriata ) was collected and identified during a mycoviral screening survey conducted in 2018 from a diseased leaf of the Lokath plant ( Khan et al., 2021b ). Some mycoviruses identified as a result of this screening survey have already been published ( Khan et al., 2021a ; Khan et al., 2021b ; Khan et al., 2022 ). For isolating purified cultures of L3, surface sterilization of leaves was performed with 1% sodium hypochlorite solution and further washing was done using autoclaved sterile distilled water.…”

“…The dsRNA patterns of 16 fungal isolates out of 36 dsRNA-positive ones are shown in , many of which show multiple bands. Several previously characterized fungal strains infected by RNA viruses were included in the dsRNA analysis as references, which included a botybirnavirus (AaBbV1)-infected Alternaria alternata isolate 4 a [34], a hadakavirus (HadV1-7n)-infected Fusarium oxysporum isolate 7 n and a HadV1-1NL infected Fusarium nygami isolate 1 NL [17, 35] (Fig. S2).…”
