A Role for the Septation Initiation Network in Septum Assembly Revealed by Genetic Analysis of sid2-250 Suppressors

Abstract: In the fission yeast Schizosaccharomyces pombe the septation initiation network (SIN) is required for stabilization of the actomyosin ring in late mitosis as well as for ring constriction and septum deposition. In a genetic screen for suppressors of the SIN mutant sid2-250, we isolated a mutation, ace2-35, in the transcription factor Ace2p. Both ace2D and ace2-35 show defects in cell separation, and both can rescue the growth defects of some SIN mutants at low restrictive temperatures, where the SIN single mut… Show more

“…Overexpression of Rho1 partially rescues lethality of Sid2 mutants at low restrictive temperature (Jin et al 2006). Interestingly, we observed that moderate overexpression of Rho1 under the nmt41x promoter in a plasmid (p41rho1) efficiently suppressed lethality of etd1D mutants ( Figure 1C), suggesting that Etd1, as proposed for Sid2 (Jin et al 2006), functions in downstream activation of Rho1.…”

“…In addition, Etd1 localization to the medial ring requires Sid2-Mob1 activity (Daga et al 2005). Thus, we further analyzed whether Etd1 is involved in connecting Sid2-mediated SIN signaling with activation of Rho1 and the cell-wall synthesis machinery.Overexpression of Rho1 partially rescues lethality of Sid2 mutants at low restrictive temperature (Jin et al 2006). “…The double null mutant is viable, but is sensitive to temperature (high and low) and stress and shows defects in septation and cleavage ( Jiang and Hallberg 2000;Tanabe et al 2001). Curiously, a mutation in the regulatory subunit B9 Par1 is able to suppress the septation defects of cdc11-136, cdc7-24, spg1-B8 (Le Goff et al 2001), and sid2-250 mutants ( Jin et al 2006).…”

“…Sid2p interacts with Mob1p and this interaction is required both for localisation of Sid2p to a medial ring where the septum will form and for Sid2p catalytic activity (Hou et al, 2004). Sid2p may promote septum formation by activating the S. pombe homologue of Ace2p, but the precise mechanism of how it does this remains unclear (Jin et al, 2006). A Cdc15-related kinase, Cdc7p, is also required for septum formation; however direct activation of Sid2p by Cdc7p has not been demonstrated (Mehta and Gould, 2006).…”
