A role for Pin1 in mammalian germ cell development and spermatogenesis

Abstract: The peptidyl-prolyl isomerase Pin1 is proposed to have diverse functions in many vital aspects of the cell. Despite the multitude of proteins targeted by Pin1 and the proposed regulatory role it plays in critical cellular functions, Pin1 is an essential gene in some eukaryotic organisms, but is dispensable in metazoans. In two genetic models, Candida albicans and Drosophila melanogaster, Pin1 participates in distinct developmental processes regulated by the MAPK pathway. Pin1-deficient mice exhibit decreased p… Show more

“…The prolyl-isomerase Pin1 is required for the self-renewal of normal mammary stem cells Pin1 knock-out mice show a number of developmental defects (Atchison & Means, 2004) affecting among others mammary epithelium, that fails to undergo the dynamic changes required to its expansion during pregnancy . Based on this, we hypothesized a possible function of Pin1 in governing the functions of mammary stem cells and thus we evaluated the stem cell activity of mammary epithelial cells from wild-type (Pin1 +/+ ) and knock-out (Pin1 À/À ) mice.…”

“…Pin1 is unique among the peptidyl-prolyl isomerases because it specifically recognizes phosphorylated serine or threonine residues immediately preceding proline (Ser(P)/Thr-Pro) in a subset of proteins and isomerizes the cis/trans conformation of the peptide bond (10,11). Several studies have shown that Pin1-mediated conformational regulation can have a profound impact on the regulation of cell growth, stress responses, immune function, germ cell development, neuronal differentiation, and survival (12,13). Dysregulation of Pin1 signaling is implicated in some pathological con-* This work was supported, in whole or in part, by National Institutes of Health ditions such as Alzheimer disease (14,15), asthma (16,17), corticobasal degeneration (18,19), and cancer (20).…”
