A role for Drosophila Wnt‐4 in heart development

Abstract: Summary:In vertebrates, different Wnt-signaling pathways are required in a temporally coordinated manner to promote cardiogenesis. In Drosophila, wingless holds an essential role in heart development. Among the known Drosophila Wnts is DWnt4, the function of which has been studied in various developmental processes except for heart development. We re-evaluated the expression pattern of DWnt4 during embryogenesis and show that transcripts are not restricted to the dorsal ectoderm but are also present in the car… Show more

“…A mutation in the Wnt4 gene ( Wnt4 EMS23 ) was identified in this large-scale screen and was classified in the category of abnormal cardiac morphology. The cardiac phenotype of Wnt4 mutants was also described in an independent study (Tauc et al, 2012), and was interpreted as an early cardiac specification defect. However, our detailed analysis of the Wnt4 mutant phenotype and its expression pattern suggest that Wnt4 is not required for cardiac specification, but required for ostia development.…”

“…A previous study of Wnt4 in the Drosophila heart did not identify downstream Wnt signaling pathway components (Tauc et al, 2012). We used multiple approaches to demonstrate that Wnt4 functions through the canonical Wnt signaling pathway.…”

“…To test this more directly, we overexpressed dWnt4 in piRNA mutants to see if it would rescue the CB differentiation phenotype. We used c587-GAL4 to drive UASt dWnt4 in dSETDB1 mutants and stained them with Vasa and 1B1 [ 47 ]. Additionally, we depleted dSETDB1 and piwi only in the somatic cells, while simultaneously expressing dWnt4 there.…”

“…A recent study described enriched expression of a Wg paralog, DWnt4 , in the ostia cells (Tauc et al 2012), and it is likely that this factor also contributes to Wg signaling in the ostia and potentially to ostia formation. Moreover, since canonical Wg signaling is not sufficient for ostia specification, factors in addition to Wg signaling are necessary for ostia formation, and our studies place these yet-to-be defined factors genetically downstream of abd-A .…”
