A Role for Drosophila Amyloid Precursor Protein in Retrograde Trafficking of L1-Type Cell Adhesion Molecule Neuroglian

Abstract: The role of the Amyloid Precursor Protein (APP) in the pathology of Alzheimer’s disease (AD) has been well studied. However, the normal function of APP in the nervous system is poorly understood. Here, we characterized the role of the Drosophila homolog (APPL) in the adult giant fiber (GF) neurons. We find that endogenous APPL is transported from the synapse to the soma in the adult. Live-imaging revealed that retrograde moving APPL vesicles co-traffic with L1-type cell adhesion molecule… Show more

“…To confirm that the distribution of APPL's ICD in secretory compartments was not grossly altered by the addition of a red fluorescent tag, we assessed the localisation of an APPL-EGFP C-terminal fusion in SCs (Figure 4A; Penserga et al, 2019). The EGFP-labelled ICD of APPL was observed at the limiting membrane of DCG compartments and their precursors (Figure 4D, boxes 2 and 1 respectively), mirroring the distribution of the mRFP-tagged ICD of double-tagged APPL; it was also more frequently and clearly detected in ILVs and at the periphery of DCGs.…”

“…In addition, Appl loss-of-function phenotypes can be rescued by human APP, suggesting functional conservation (Luo et al, 1992). Although the specific molecular and subcellular functions of APPL remain unclear, the protein has been reported to control the balance between regulated secretion and retrograde transport at the synapse (Penserga et al, 2019).…”

“…In neurons, APP is transported along axons via kinesin-dependent anterograde transport and accumulates in synaptic sites [ 93 , 94 ]. In Drosophila , APP-like (APPL) was found to regulate the sorting of the adhesion molecule neuroglian, which has a role in synaptogenesis by organizing cytoskeletal molecules in synaptic terminals [ 95 , 96 ].…”

“…Furthermore, Nrg was implicated in the remodeling of the Drosophila Giant Fiber (GF) circuitry, which exhibits pruning of extraneous axonal branches during the pupal stage ( Borgen et al, 2017 ). In this system, Nrg was shown to be retrogradely transported from GF terminals in an Amyloid Precursor Protein-like (APPL)-dependent manner ( Kudumala et al, 2017 ; Penserga et al, 2019 ). APPL mutants exhibit pruning defects of GF transient branches, thus implying a potential role for Nrg in GF pruning, although this was not directly tested.…”
