A rhesus macaque model of Asian-lineage Zika virus infection

Abstract: Infection with Asian-lineage Zika virus (ZIKV) has been associated with Guillain–Barré syndrome and fetal abnormalities, but the underlying mechanisms remain poorly understood. Animal models of infection are thus urgently needed. Here we show that rhesus macaques are susceptible to infection by an Asian-lineage ZIKV closely related to strains currently circulating in the Americas. Following subcutaneous inoculation, ZIKV RNA is detected in plasma 1 day post infection (d.p.i.) in all animals (N=8, including 2 p… Show more

“…To assess differences in ZIKV replication between virus delivery by needle 73 versus mosquito vector, we infected rhesus macaques with the Puerto Rican ZIKV isolate 74 PRVABC59 by either sc inoculation (n=3) or by exposure to infected mosquitoes (n=4). All three 75 sc-inoculated macaques were productively infected, with viral load dynamics similar to what we 76 have reported previously 1,2 . All four animals exposed to Ae.…”

“…Our studies using the same route and doses ranging from 1 x 10 4 to 1 x 10 6 60 PFU of a French Polynesian isolate showed no persistence in body fluids after the resolution of 61 acute plasma viremia in non-pregnant macaques 2 . In yet another study in rhesus macaques, 62 intravenous administration of 1 x 10 5 PFU of a Brazilian ZIKV isolate resulted in a short-lived 63 plasma viremia and vRNA distribution in a variety of tissues two weeks post-infection 7 .…”

“…[34][35][36][37] Li et al 38 observed that experimental infection of infected embryonic mouse brains with ZIKV resulted in smaller brain size with enlarged lateral ventricles and thinner cortical plates when compared with uninfected controls. In this model, the neural progenitor cells and intermediate or basal progenitor cells were the primary cells affected.…”
