Design and Characterization of a Natural Arf-GEFs Inhibitor Prodrug CHNQD-01255 with Potent Anti-Hepatocellular Carcinoma Efficacy In Vivo

Brefeldin A (BFA), a well-known natural Arf-GEFs inhibitor, is effective against hepatocellular carcinoma (HCC), while the poor solubility, serious toxicity, and short half-life limit its potential. Herein, distinct corresponding prodrugs of BFA, including esters 1–15, carbonates 16–24 and 30–32, and carbamates 25–29, were synthesized and evaluated. CHNQD-01255 (16) with improved aqueous solubility (15–20 mg/mL) demonstrated favorable pharmacokinetic profiles. It behaved as expected by undergoing rapid conversion to BFA in vivo, and achieved sufficient high plasma exposure, prolonged half-life, as well as the improved bioavailability of BFA (F = 18.96%). Meanwhile, CHNQD-01255 significantly suppressed tumor growth (TGI = 61.0%) at a dose of 45 mg/kg (p.o.) in the xenograft model. Notably, the improved safety profile of CHNQD-01255 (MTD > 750 mg/kg, p.o.) was confirmed to be superior to that of BFA (MTD < 506 mg/kg). Overall, CHNQD-01255 may serve as a safe and effective new anti-HCC prodrug.

A study was carried out to look into the effects of dietary zinc and nicotinic acid (niacin) supplementation on growth performance, hematology and biochemical parameters of turkey. The birds were distributed in to four treatments each treatment have three replicates. (3 birds / replicate) T1 no additive as control, fed diets supplemented with Zn (100 mg/kg) T2, Niacin (100 mg/kg diet) T3 and combination of them Zn100 mg/kg+ niacin 100 mg/kg) T4. the results analyzed by CRD arrangement design. Ayam broiler adalah jenis ayam yang dipelihara khusus untuk produksi daging yang cepat, sehingga pemeliharaan yang baik sangat penting untuk mencapai performa yang optimal. Kunci pemeliharaan ayam broiler dengan skala produksi adalah pakan, manajemen kandang, manajemen kesehatan, dan faktor eksternal. Perlu dikembangkan cara-cara yang baru dalam pemeliharaan ayam broiler agar menghasilkan kualitas yang baik. Salah satunya adalah dengan mengem-bangkan produk suplemen pendamping pakan. Sebanyak 12.000 ekor ayam DOC dibagi kedalam 2 kelompok yai-tu kelompok kontrol yang diberikan pemeliharaan secara normal. Kelompok berikutnya dalah kelompok EBS yaitu kelompok yang diberikan produk Elektro Bio Stamino® setiap hari didalam air minum dengan dosis 2 mL dalam 1 liter air minum. Pemberian sediaan dilakukan sebanyak 1 kali sehari. Parameter yang diamati meliputi penambahan bobot badan, Feed consumption rate dan Feed intake. Pengambilan data dilakukan selama 31 hari. Hasil penelitian menunjuk-kan kelompok yang diberikan sediaan Elektro Bio Stamino® memiliki performa yang lebih baik dibandingkan dengan kelompok kontrol. Hal ini disebabkan nutrisi dan zat yang terkandung didalam produk mampu melengkapi kebutu-han nutrisi ayam selama masa pemeliharaan. Oleh karena itu pemberian Elektro Bio Stamino® memberikan efek positif terhadap performa ayam broiler.
