A review of mechanistic insight and application of pH-sensitive liposomes in drug delivery

Abstract: The pH-sensitive liposomes have been extensively used as an alternative to conventional liposomes in effective intracellular delivery of therapeutics/antigen/DNA/diagnostics to various compartments of the target cell. Such liposomes are destabilized under acidic conditions of the endocytotic pathway as they usually contain pH-sensitive lipid components. Therefore, the encapsulated content is delivered into the intracellular bio-environment through destabilization or its fusion with the endosomal membrane. The … Show more

“…Different strategies based on liposomal vesicles were designed with the intention of streamlining these functions: examples include acidic pH-sensitive liposomes (Lp) (6, 7), cationic Lp (8, 9), and the inclusion of immunomodulators, such as TLR ligands (10, 11). Liposomal preparations including membranotropic peptides and bacteria pore-forming proteins (PFPs) also were investigated as novel cytosol-delivery systems for cross-priming cytotoxic CD8 + T cells (12, 13).…”

“…(El-Badry et al, 2014). Liposomes are also effective in loading of water-soluble drugs in central aqueous compartment and lipid soluble agents in outer membrane (Paliwal et al, 2015). Due to the advancements in the methods of preparing and formulating liposomes, high-entrapment efficiencies can be made possible to incorporate different types of drugs into them (Kulkarni et al, 2011).…”

“…Once inside, the drug can be remotely offloaded via the endosomal pathway by hydrolytic cleavage or rapid degradation of the drug carrier triggered by an acidic environment [18,19]. Our group is one of the first to demonstrate the successful targeting of cancer cell-internalized pathogenic bacteria.…”
