A Residence-Time Distribution Analysis of the Hydrodynamics within the Intestine in Man during a Regional Single-pass Perfusion with Loc-I-Gut: In-vivo Permeability Estimation

Abstract: The goal of this study was to determine the most appropriate hydrodynamic model for the Loc‐I‐Gut in‐vivo perfusion system. The general mixing‐tank‐in‐series model, which can approximate single mixing tank and laminar and plug‐flow hydrodynamics, was fitted to the observed experimental residence‐time distribution curves for the non‐absorbable marker [14C]PEG 4000. The residence‐time distribution analysis shows that the hydrodynamics of the perfusion solution within the jejunal segment in man is well approximat… Show more

“…1). a BCS classification from reference (4) b Observed f abs data from reference (33) Despite this simplification, P eff , as it is, has been an effective predictor of both the rate and extent the GI absorption (2,16,61). This discrepancy can be explained by the following differential equation that describes the drug absorption from the SI (assuming the SI as single compartment) (1,2):…”

“…When the perfusate (NuTRIflex) reached the intestinal lumen it was immediately diluted according to the well-mixed hydrodynamics (28). The concentrations of the lipidic components in the NuTRIflex solution upon entry in the jejunum were therefore calculated for each time interval under the assumption that dilution took place immediately upon entering the small intestine and that no degradation or absorption occurred.…”
