A Requirement for Flk1 in Primitive and Definitive Hematopoiesis and Vasculogenesis

Abstract: Mouse embryos lacking the receptor tyrosine kinase, Flk1, die without mature endothelial and hematopoietic cells. To investigate the role of Flk1 during vasculogenesis and hematopoiesis, we examined the developmental potential of Flk1-/- embryonic stem cells in chimeras. We show that Flk1 is required cell autonomously for endothelial development. Furthermore, Flk1-/- cells do not contribute to primitive hematopoiesis in chimeric yolk sacs or definitive hematopoiesis in adult chimeras and chimeric fetal livers.… Show more

“…VEGFR-1 mediates macrophage chemotaxis (Barleon et al, 1996) and the organization of the embryo vasculature by vasculogenesis (Fong et al, 1995), but not the definitive vessel assembly, which is closely dependent on VEGFR-2, a specific EC differentiation marker (Shalaby et al, 1999). The bFGF/ FGFRs system is mandatory for vasculogenesis, since it induces angioblasts to differentiate and assemble into initial vascular patterns (Gendron et al, 1996).…”

“…VEGF and VEGFR2 play a pivotal role in maintaining EC integrity as demonstrated by targeted disruption of these genes. 3,36,37 The antiapoptotic effect of VEGF and also of angiopoietin-1 (Ang-1; via activation of its receptor Tie2) is mainly mediated through activation of the survival-promoting PI3K/Akt signaling pathway. 38,39 By diverse mechanisms, the activation of PI3K/Akt inhibits the apoptosis of ECs [40][41][42][43] VEGF upregulates antiapoptotic proteins such as survivin, XIAP and Bcl-2.…”

“…VEGF-R2/KDR is required for vasculogenesis and hematopoiesis (Eichmann et al, 1997;Shalaby et al, 1997). Thus, prolonged inhibition of VEGF-R signaling could inhibit hematopoiesis in zebrafish embryos, and thereby affect blood viscosity and shear forces in the developing vasculature.…”
