Induction of 2n pollen is a required technique for cultivating polyploid via sexual polyploidy. Orthogonal design or Taguchi Design was applied to select the best treatment process of 2n pollen induction inPopulus×popularisfrom different levels of the meiosis stage of male flower buds, colchicine concentration, times of injection, and interval between injections. Flow cytometry and chromosome counting were used to identify the triploids from the offspring ofP.×euramericana. (Dode) Guinier pollinated with induced pollen ofP.×popularis. The results showed that high 2n pollen rate can be achieved by selecting the flower buds during diakinesis stage in meiosis, and then injecting 0.6% colchicine 4 times with 2 hours interval. The 2n pollen rate reached 62.10% by this process, and two triploids were obtained, which indicates that it is possible for cultivating triploids via 2n pollen induction by colchicine treatment in poplar. Results and protocol related to 2n pollen induction, polyploid identification and effect of 2n pollen in this study might be applicable in polyploidy breeding in sectionAigeirosandTacamahacaof poplar.

Unreduced gametes are the driving force for the polyploidizaiton of plants in nature, and are also an important tool for breeding of triploid individuals. The final heterozygosity of a 2n pollen grain depends on the cytological mechanism behind 2n pollen formation. In this study, meiotic abnormalities were analysed using fluorescent chromosome staining and indirect immunofluorescence during the microsporogenesis of 18 genotypes of diploid P. euphratica Oliv. (2n = 2x = 38). Among the 18 genotypes, 16 genotypes produce 2n pollen and two genotypes produce only normal n pollen. In all 2n pollen producers, we found that the first meiotic division was normal but that the second division was characterized by frequent abnormal spindle orientation (parallel, tripolar, and fused spindles) and premature cytokinesis. The parallel, fused spindles and premature cytokinesis were considered to be leading dyad formation, and tripolar spindles seemed to be causing triad formation at the tetrad stage. There was a higher frequency of parallel spindles than other spindle forms, but no significant correlations between parallel spindles and dyads were observed. However, a significant association (r = 0.68, P < 0.05) between the tripolar spindles and dyads was found. In some Microspore mother cells (MMCs), an indirect immunofluorescence examination of meiosis II revealed that the parallel spindles led to the gathering of one or two non-sister groups of chromosomes, causing an incorporation of RMSs from two daughter nuclei. Therefore, the incorporated RMSs established two nuclear cytoplasmic domains for the control of division plane, resulting in either triad or dyad formation.

The most direct approach in breeding triploid Populus is crossing allotetraploids with diploids. However, the lack of allotetraploid Populus restricts application of this approach. In this investigation, zygotic chromosome doubling was induced with colchicine and high temperatures to produce ((Populus pseudo-simonii × P. nigra 'Zheyin3#') × (P. × beijingensis)) allotetraploids. We screened 6 and 25 tetraploid individual offspring from the colchicine and high-temperature treatments respectively, indicating that both colchicine and high temperature are effective for tetraploid production by zygotic chromosome doubling of Populus. Developmental characteristics of seed hairs in the ovaries were temporally associated with zygotic development, which was used to successfully guide the colchicine and high-temperature treatments. During certain stages of hair development, the efficiency of tetraploid production was significantly high. However, efficiency of production was not significantly influenced by other factors, i.e. colchicine concentration, temperature or duration of high-temperature treatment. Size and frequency of leaf stomata between tetraploid and diploid plants were significantly different, suggesting that this character can be altered via genomic increase in material. The allotetraploids produced in this investigation, having different genotypes, provide important parental germplasms for further triploid breeding.
