A rainbow trout SRY‐type gene expressed in pituitary glands

Abstract: A Sox (SRY-type HMG box) gene, designated SoxP1, was isolated from a cDNA library made from pituitaries of immature rainbow trout. Sequence analysis indicated that the cDNA had an open reading frame encoding 467 amino acid residues containing a DNA binding motif, known as the high mobility group (HMG) box. Northern blot analysis showed trout SoxP1 mRNA was detected in pituitaries and gonadal tissues, but not in liver, spleen, and heart. In pituitaries, trout SoxP1 mRNA was more abundant in immature fish than i… Show more

“…Recombinant SOX13 was tested for binding to radiolabeled double-stranded DNA bearing the high-affinity SOX binding motif AACAAT (29,34,35). SOX13(66-604) protein bound the AACAAT sequence but not the CCGCGG sequence ( F i g .…”

“…Sox10 belongs to the same subgroup of Sox proteins as Sox8 and Sox9, as indicated by the fact that the HMG domains of all three proteins share Ͼ90% identity (Wright et al, 1993). Sequence similarities were apparent even outside the (Wright et al, 1995), mouse Sox8 (Wright et al, 1993), and rainbow trout SoxP1 (Ito et al, 1995). E xact matches between Sox10 and the aligned sequences are marked by asterisks.…”

“…1C). When amino acid sequences of rat Sox10 and mouse Sox9 (Wright et al, 1995) were aligned with sequences of SoxP1, the putative Sox8 ortholog from rainbow trout (Ito et al, 1995), we detected three additional regions with significant sequence conservation. One of them corresponded to amino acids 233-306 of Sox10 with 61% identity over a stretch of 74 amino acids, and a second one corresponded to the C -terminal region of Sox10 with 41% identity over a stretch of 66 amino acids.…”

“…The SRY gene belongs to a rapidly growing family of genes that are related by sequence homology to the HMG box, named Sox genes (SRY-like HMG-box gene). The members of the Sox gene family have been conserved through evolution, and have been found in a wide variety of species including humans [7], mice [36], marsupials [11], birds [14], turtles [26], Xenopus [23], alligators, lizards, Drosophila [6] and fishes [12,15,29,30,32]. Although recent studies show that some Sox genes have important developmental roles, many of them have not been identified.…”
