A psychophysical and electrophysiological analysis of salt taste in Trpv1 null mice

Abstract: Current evidence suggests salt taste transduction involves at least two mechanisms, one that is amiloride sensitive and appears to use apically located epithelial sodium channels relatively selective for Na(+) and a second that is amiloride insensitive and uses a variant of the transient receptor potential vanilloid receptor 1 (TRPV1) that serves as a nonspecific cation channel. To provide a functional context for these findings, we trained Trpv1 knockout (KO) and wild-type (WT) C57BL/6J mice (n = 9 or 10/grou… Show more

“…This such as CAP and piperine, are received by the transient receptor potential cation channels known as subfamily V, member 1 (TRPV1) (Tominaga et al, 1998;Okumura et al, 2010). TRPV1 variant is reported as a candidate molecule for the salt receptor (Lyall et al, 2004;Treesukosol et al, 2007). These receptors may affect the enhancement of salt taste intensity by CAP.…”

“…Control stimuli consisting of 300 mM NH 4Cl and 300 mM NaCl applied at the beginning and end of the experiment were used to assess preparation stability (31,32). As in our previous studies, the control responses did not differ by more than 2-5% at the beginning and end of the experiment (11,48). The stimulus series used in the CT experiments to generate the relationship between RTX concentration and the magnitude of the NaCl ϩ Bz CT response was as follows: R ¡ 300 mM NH 4Cl ¡ R ¡ 300 mM NaCl ¡ R ¡ 100 mM NaCl ¡ R ¡ (100 mM NaCl…”

“…These mice have a targeted mutation caused by a disrupted exon encoding part of the fifth and all of the sixth transmembrane pore regions; age-matched C57 mice, which we obtained from Charles River Laboratories are the appropriate control mice (Caterina et al, 2000;Ciura and Bourque, 2006;Treesukosol et al, 2007). Mice were maintained in a 12 h light/dark cycle with standard rodent chow available ad libitum.…”
