Due to its ornamental and medicinal value, pear flower has been historically loved and used in China. However, the current understanding of their odor-active compounds and aroma profiles is rather limited. This work aimed to evaluate and compare the overall aroma profile of pear flowers; the volatiles in flowers of seven pear cultivars (Anli, Bayuesu, Golden, Brown peel, KorlaXiangli, Lyubaoshi, Xizilü) were analyzed using solid-phase microextraction–gas chromatography-mass spectrometry (SPME-GC-MS). A total of 93 volatile compounds were identified and quantified within the amount of volatiles in the range of 62.7–691.8 μg kg−1 (FW) and showed high and significant variability in different cultivars. Anli and Brown peel flowers showed a relatively higher volatile abundance, while KorlaXiangli flowers were significantly lower than other cultivars. Although the composition of volatiles depended on the existence of different chemical classes, the odor activity values (OAVs) and odor descriptions showed some aldehydes were part of their main peculiarities and were considered as the basic active odorants that presented strong intensity of citrus and floral odor. Moreover, multivariate analysis showed the pear flower of different cultivars could be arranged in different clusters by the identified odorants. This study provides first-hand knowledge regarding pear flower aroma profiles, and that the cultivar differences were critical for the overall pattern.

Structural Diversity of Streptococcal Mutans Synthesized under Different Culture and Environmental Conditions and Its Effect on Mutanase Synthesis

Streptococcal mutans synthesized under different conditions by growing cultures or by their glucosyltransferases were shown to exhibit a great structural and property diversity. Culturing and environmental factors causing structural differences in mutans were specified. All of the obtained biopolymers (76 samples) were water-insoluble and most of them (72) had a structure with a predominance of α-(1→3)-linked glucose (i.e., the content of α-(1→3)-linkages in the glucan was always higher than 50%, but did not exceed 76%). An exception were four glucans containing more than 50% of α-(1→6)-sequences. In these structurally unique mutans, the ratio of α-(1→3)- to α-(1→6)-bonds ranged from 0.75 to 0.97. Aside from one polymer, all others had a heavily branched structures and differed in the number of α-(1→3), α-(1→6), and α-(1→3,6) linkages and their mutual proportion. The induction of mutanase production in shaken flask cultures of Trichoderma harzianum by the structurally diverse mutans resulted in enzyme activities ranging from 0.144 to 1.051 U/mL. No statistical correlation was found between the total percentage content of α-(1→3)-linkages in the α-glucan and mutanase activity. Thus, despite biosynthetic differences causing structural variation in the mutans, it did not matter which mutan structures were used to induce mutanase production.

Effects of mutations at threonine-654 on the insoluble glucan synthesized by Leuconostoc mesenteroides NRRL B-1118 glucansucrase

Twelve different amino acids were each substituted for threonine-654 in a cloned glucansucrase from Leuconostoc mesenteroides NRRL B-1118. Both the native and the cloned enzyme with threonine at position 654 produced a water-insoluble glucan containing approximately 44 mol% 1,3-disubstituted α-D-glucopyranosyl units and 29 mol% 1,6-disubstituted α-D-glucopyranosyl units. Several substitutions yielded an enzyme that produced an increased percentage of 1,3-disubstituted α-D-glucopyranosyl units, with corresponding decreases in 1,6-disubstituted α-D-glucopyranosyl units. Only one substitution, tyrosine, resulted in a significant increase in the percentage of 1,6-disubstituted α-D-glucopyranosyl units, with a concomitant increase in glucan yield. The mutated enzymes that produced the highest levels of 1,3-disubstituted α-D-glucopyranosyl units were also significantly activated by the addition of dextran, but glucan yields were also lower in these mutants.
