A prevascularized subcutaneous device-less site for islet and cellular transplantation

Abstract: Transplantation of donor-derived islets into the liver is a successful cellular replacement therapy for individuals with diabetes. However, the hepatic vasculature is not an optimal transplant site for several reasons, including graft attrition and the inability to retrieve or image the islets. Here we describe islet transplantation into a prevascularized, subcutaneous site created by temporary placement of a medically approved vascular access catheter. In mice with streptozotocin (STZ)-induced diabetes, trans… Show more

“…Inducing dense angiogenesis at close proximity to the graft capsule may create a more supportive environment. Such induction attempts included temporal placement of proan giogenic membrane or mesh [195,196] , slow release of proangiogenic factors [197200] , and using both these strategies concurrently [201] . Enhanced angiogenesis that promoted longterm islet function occurred, but was validated only in rodent models.…”

Ces chercheurs observent un effet bénéfique de ce système sur la fonctionnalité des îlots greffés et sur leur capacité à normaliser les glycémies chez des souris diabétiques transplantées.…”

“…The DL space was created as previously reported by inserting a nylon catheter subcutaneously in the left lower abdomen and left for five weeks before transplant [9]. Diabetes was chemically induced by intraperitoneally injecting 180 mg/kg of streptozotocin (STZ; Sigma-Aldrich, ON, Canada) in half of the recipients, one week prior to transplantation.…”

“…Our group recently described a novel pre-vascularized DeviceLess (DL) technique for cell transplantation in the subcutaneous space [9]. This approach was successful in reversing diabetes with mouse and human islets and is currently being used for other cell therapies.…”
