A preliminary report: genistein attenuates cerebral ischemia injury in ovariectomized rats via regulation of the PI3K-Akt-mTOR pathway

Abstract: Stroke is a leading cause of disability and death in the worldwide. Therefore, prevention of stroke is critically important. Genistein, a natural phytoestrogen extracted from soybeans, has been found to be a potential neuroprotective agent for stroke prevention. However, the role of genistein and its underlying mechanism in ovariectomized rats has been rarely evaluated. In this study, ovariectomized rats were treated with genistein (10 mg/kg) or vehicle daily for two weeks before they received middle cerebral … Show more

“…Isoflavones. In total, 6 isoflavones are found to possess neuroprotective activity in ischemic stroke, that is, calycosin [43,70,190,191], calycosin-7-O-β-D-glucoside [192], formononetin [193], genistein [194][195][196][197][198][199], daidzein [200], and puerarin [103,110,[201][202][203].…”

“…It was found that genistein pretreatment markedly decreased the neurological deficits and infarct volumes of ovariectomized MCAO rodents. The mechanisms involved the promotion of Nrf2-mediated antioxidation and inhibition of apoptosis by activating the PI3K/Akt/mTOR pathway [197][198][199]. Besides, equol, a metabolite of daidzein, was found to exert neuroprotection in the ovariectomized MCAO/R rats by enhancing the antioxidant defense [206].…”

“…Genistein can also directly act on vascular endothelial cells, alleviating oxidative stress, in ammation and vascular damage by increasing the protein expression of eNOS [67] . At present, the research on Genistein mainly focuses on ischemic stroke, brain injury and Alzheimer's disease [68,69] , with few reports on epilepsy. In this experiment, we observed that activated microglia and astrocytes, expression of JAK2/STAT3 in ammation pathway, and apoptotic caspase3 and Bax increased signi cantly after epilepsy, while the number of neurons, expression of Nrf2 anti-oxidative stress pathway and anti-apoptotic protein Bcl-2 were reduced.…”

“…The role of the PI3K/AKT pathway in stroke is well-known. It regulates downstream anti-apoptotic molecules and plays a protective role in rats after ischaemic stroke ( Li, Liu, et al., 2019 ; Lu et al., 2019 ; Meng et al., 2020; Wei et al., 2020 ; Yang et al., 2020 ). However, PI3K/AKT has not been associated with the NLRP3 inflammasome in cerebrovascular diseases.…”
