The effect of application timing of metconazole on the control of Fusarium head blight (HFB) and mycotoxin accumulation in wheat and barley was investigated. Single and double applications of metconazole at different spray timings were performed in a wheat field trial. In single application plots, the mid-flowering stage was the optimal application timing of metconazole for controlling FHB symptom development, and the milking stage was optimal for reducing mycotoxin contamination. Similar efficacies were observed in all spray timings in double application plots, which included the mid-flowering stage and another spray timing. In the barley field trial, application at the mid-flowering stage controlled both FHB development and mycotoxin contamination with the highest efficacy. Similar efficacies for controlling the development of FHB in the barley field were observed with single and double applications of the fungicide. However, mycotoxin contamination was lower with double application than with single application of the fungicide.

A rapid and sensitive method based on competitive indirect enzyme-linked immunosorbent assay (ciELISA) was developed and validated for the detection of DON (deoxynivalenol) in cereals with the confirmation of the reliable ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/ MS). Within this method, the IC50 (half maximal inhibitory concentration of a substance) of the DON-specific monoclonal antibody (MAb) we produced was 61.10 ng/mL. The limit of detection (LOD) value, measured by IC10, was 6.12 ng/ mL, which is below the maximum residue levels (MRLs) established for DON in various cereals. In spiked samples, the recoveries ranged from 79.3% to 115.2% for maize and from 74.3% to 118.3% for wheat. This method was compared with the UPLC-MS/MS method by testing four concentrations. The correlation coefficient for the two methods was 0.9884 in a linear-regression analysis. The results illustrated the reliability of the ciELISA method for the determination of cereal samples.

Through an analysis of T-2 and HT-2 toxins in corn and brown rice, the effect of enzymatic deacetylation of T-2 toxin on HT-2 toxin was investigated. Gas chromatography (GC) with electron capture detection and high-performance liquid chromatography (HPLC) with fluorescence detection were used for quantitative determination. T-2 toxin was converted into HT-2 (84-86%) within 15 min in the presence of crude protein extracts from corn and brown rice. The absence of T-2 conversion was observed for autoclaved samples, in which the enzymes were inactivated. When phosphate buffered saline, followed by methanol, was used as the extraction solvent, recoveries of T-2 toxin spiked at 50 and 200 µg/kg were from 60 to 87%, whereas those of HT-2 in the autoclaved samples were 0%. In non-autoclaved samples, recoveries of HT-2 were 37-66%, whereas those of T-2 were negligible. However, the conversion of T-2 into HT-2 was not observed when samples were extracted by methanol/water.
