A Population of Murine γδ T Cells That Recognize an Inducible MHC Class Ib Molecule

Abstract: Although gammadelta T cells are implicated in regulating immune responses, gammadelta T cell-ligand pairs that could mediate such regulatory functions have not been identified. Here, the expression of the major histocompatibility complex (MHC) class Ib T22 and the closely related T10 molecules is shown to be activation-induced, and they confer specificity to about 0.4% of the gammadelta T cells in normal mice. Thus, the increased expression of T22 and/or T10 might trigger immunoregulatory gammadelta T cells du… Show more

“…This finding is consistent with other recent studies associating TCR-defined subsets of ␥␦ T cells with distinct functional roles (23,(25)(26)(27)(28). The requirement for a TCR-defined population also suggests that TCR-ligand recognition and the particular specific ligand of the regulatory ␥␦ T cell subset are important in maintaining normal airway function (29).…”

“…5B). T10 and T22 are two closely homologous nonclassical MHC molecules that act as ligands for roughly 0.5% of all ␥␦ T cells in many inbred mouse strains (33). Increased expression of these nonclassical MHC molecules was not due simply to an up-regulation of all MHCI like molecules caused by increased ␤ 2 -microglobulin expression (Table I).…”

“…Thermal denaturation studies have been performed with one additional nonclassical class I molecule, T10 (40). T10 and the closely related T22 (94% identity) are recognized by a subpopulation of ␥␦ T cells in mice without a requirement for bound peptide or other components (40,43). The crystal structure of T22 demonstrates a severely modified FIGURE 4.…”
