A Personal View on the Early History of the Insulin-Like Growth Factors

Abstract: Salmon and Daughaday, when trying to set up an in vitro assay for Growth Hormone (GH), failed to obtain a direct effect on sulphate uptake in cartilage of hypophysectomized (hypox) rats. They recognised that this was not the consequence of poor methodology or materials, but an encrypted message from the examined system. They decided to turn around and to try and decipher it. Treatment with GH appeared to render hypox rat serum active in stimulating sulphation in hypox rat cartilage. They proposed that GH induc… Show more

“…These observations support early evidence that IGF-I mediates the stimulatory effect of growth hormone on longitudinal bone growth during development (Van den Brande, 1999;Sims et al, 2000). Subsequent studies revealed that certain hormones have important osteogenic effects that depend at least in part on changes in IGF-I synthesis.…”

“…However, APOE genotype did not affect the length of life after diagnosis. Some studies found that the presence of at least one APOE ε4 allele implied an earlier onset [36] and a faster rate of cognitive deterioration [19,37]. However, other authors reported no difference in the onset of AD [38] and progression rate between ε4 carriers and noncarriers [38,39], or a faster cognitive decline in noncarriers [40].…”

“…**** Inorganic sulfate can be used; however, most of the sulfate in the body is derived from catabolism of the amino acids *** This presumes that the nutritional requirements for hormone and growth factor formation are being met for normal lean tissue synthesis, in particular the requirements of vitamin D, iodine, and essential fatty acids. **** In animal studies, bone growth is measured by the incorporation of radioactive sulfur into skeletal tissue [68,69]. These assays, when used for factors in blood that stimulate bone growth, show low levels of these factors in malnourished children [70].…”
