A peroxidase-coupled method for the colorimetric determination of serum triglycerides.

Abstract: We describe an enzymatic method for rapid, precise measurement of serum triglycerides with use of sample:reagent ratios as large as 1:200. Hydrolysis of triglycerides is catalyzed by lipase to produce glycerol and free fatty acids. The glycerol generated is then phosphorylated by adenosine 5'-triphosphate in the presence of glycerol kinase. Oxidation of the resulting glycerol 3-phosphate to produce hydrogen peroxide is catalyzed by L-alpha-glycerophosphate oxidase. An intense red chromogen is produced by the p… Show more

“…Total cholesterol (T-chol) was analysed by a cholesterol oxidase method (Boerhinger-Mannheim, Mannheim, Germany) and triacylglyceride (Tag) by a lipase-glycerol kinase method (Sigma, Poole, Dorset, UK) with no free glycerol correction [8,9]. Both analyses were carried out on a Cobas Fara analyser (Roche Diagnostic Systems, Basel).…”

“…HDLcholesterol was measured with the same method after precipitation of very low-density and low-density lipoprotein with phosphotungstic acid and magnesium ions [18]. TG was assayed using an enzymatic method [19]. HbA 1c was measured using an automated HPLC analyser (Diamat BioRed Laboratories, NY, USA).…”

“…Triglyceride content of a sample was determined using the triglycerides lipase/peroxidase method (Fossati & Prencipe 1982;McGowan et al 1983) closely following the protocol provided by the Infinity Triglyceride kit of Thermo Electron. Two point five microlitre of supernatant was added to 125 lL of triglyceride reagent, and the absorbance measured at 520 nm after 10 min of incubation at room temperature in the dark.…”
