A pathway model of glucose-stimulated insulin secretion in the pancreatic β-cell

Abstract: The pancreas plays a critical role in maintaining glucose homeostasis through the secretion of hormones from the islets of Langerhans. Glucose-stimulated insulin secretion (GSIS) by the pancreatic β-cell is the main mechanism for reducing elevated plasma glucose. Here we present a systematic modeling workflow for the development of kinetic pathway models using the Systems Biology Markup Language (SBML). Steps include retrieval of information from databases, curation of experimental and clinical data for model … Show more

“…The pancreatic secretion pathway involves pancreatic duct cells secreting fluids and HCO 3 − . CFTR Cl − channels and CFTR-dependent Cl − /HCO 3 − ion exchange channels induced by cAMP are responsible for HCO 3 − secretion ( 24 ). The pathways enriched in the KEGG enrichment analysis mainly function in various ions’ secretion and transport—aligning with the role of ion transport in maintaining ion balance within and outside cells and fluid homeostasis in the lungs.…”

“…The action of vanadium compounds used in this study has been shown to be through PTP inhibition [9]; however, there is still the possibility of other mechanisms acting in parallel with the PTP inhibition mechanisms. As an additional biochemical study, we embarked on the investigation of the effect of the oxidovanadium(IV) compounds on glucose-stimulated insulin secretion and a stimulatory index was calculated [11,12]. From the standard curve generated (Figure S40), the insulin content of each unknown sample was determined.…”

“…In the pancreas, GK serves as a glucose sensor in β-cells, directly regulating insulin secretion [ 21 ]. As blood glucose levels rise, the increased kinetic activity of GK mediates G6P production, subsequently stimulating insulin release and maintaining glucose homeostasis [ 22 ]. In pancreatic α-cells, GK regulates glucagon secretion [ 23 ].…”
