A PARP1-ERK2 synergism is required for the induction of LTP

Abstract: Unexpectedly, a post-translational modification of DNA-binding proteins, initiating the cell response to single-strand DNA damage, was also required for long-term memory acquisition in a variety of learning paradigms. Our findings disclose a molecular mechanism based on PARP1-Erk synergism, which may underlie this phenomenon. A stimulation induced PARP1 binding to phosphorylated Erk2 in the chromatin of cerebral neurons caused Erk-induced PARP1 activation, rendering transcription factors and promoters of immed… Show more

“…Recent evidence shows inhibition of PARP1 causes loss of LTP induction in rodents [145,146]. Disruption of PARP1-ERK2 protein-protein interactions in the nucleus were responsible for this loss [145,146]. Our bioinformatic analyses suggests that synaptic protein-protein interactions with CaMKIIα, CaMKIIβ, PRKCG, or NSF may be disrupted by PARP1…”

“…PARP inhibitors have been shown to interfere with the formation of long-term potentiation (LTP) and can disrupt long-term memory formation in Aplysia bathed in inhibitors ( Cohen-Armon et al, 2004 ) and in mice when inhibitors are infused into the cerebral ventricles ( Goldberg et al, 2009 ). PARP knockout mice also demonstrate defects in LTP formation ( Visochek et al, 2016 ). It is difficult to compare the levels of PARP inhibition in these studies with those achieved by doses of PARP inhibitors administered subcutaneously or intraperitoneally to rats in the present study.…”

“…41 In the mouse hippocampus, an increase in nuclear Poly(ADP-ribose) polymerase-1 and Poly(ADP) ribose molecules during the Foskolininduced LTP is regulated by the activation of cAMPdependent protein kinase (PKA) and ERK. [42][43][44]…”
