A One-Step Immunostaining Method to Visualize Rodent Muscle Fiber Type within a Single Specimen

Abstract: In this study, we present a quadruple immunostaining method for rapid muscle fiber typing of mice and rats using antibodies specific to the adult myosin heavy chain (MyHC) isoforms MyHC1, 2A, 2X, and 2B, which are common marker proteins of distinct muscle fiber types. We developed rat monoclonal antibodies specific to each MyHC isoform and conjugated these four antibodies to fluorophores with distinct excitation and emission wavelengths. By mixing the four types of conjugated antibodies, MyHC1, 2A, 2X, and 2B … Show more

“…Muscle is composed of two types of muscle fibers, slow and fast, which have different metabolic characteristics, and these fibers are heterogeneously localized in the muscle. In gastrocnemius, we could divide it into three portions depends on the different ratio of these fibers 3 . Muscle atrophy and hypertrophy have a tendency to occur predominantly in fast fibers.…”

“…Classically, myofiber characteristics are determined based on a visual assessment of MyHC expression, and are often categorized on basis of the expression of a single and most dominant isoform only (7,(9)(10)(11). In addition, visual assessments are subjective the number of myofibers that can be scored is limited.…”
